Note:
Weight accurately approx. 100mg and add in 20ml of methanol/sulfuric acid/water (8:1:1). Dissolve, dilute with methanol to exactly 50ml. Stopper tightly and heat for one hour at 100 deg. C. Cool and filter this solution(=the Sample Solution).
Weigh accurately approx. each 2mg of Daidzein and Genistein (supplied by SIGMA CHEMICAL COMPANY), dissolve in 50ml of methanol(=the Standard solution).

Agaricus originates from Piedade in the suburbs of Sao Paulo, Brazil. About 3 decades ago, one American researcher got aware of the fact that the inhabitants in this area were rarely inflicted by adult diseases and they eat Agaricus on daily diet. It was called "Cogmelo de Deus" locally which means "God's mushroom". It was the same time that Agaricus was firstly brought into Japan by Dr. Shoji Shibata, a professor in the Pharmacological Dept of Tokyo University. Another professor named Dr. Tetsuo Ikegawa of the National Cancer Center jointly worked with Dr. Shibata on the pharmacological effects of Agaricus. The research findings were announced at the General Convention of the Japan Pharmacological Association and the Japan Cancer Association. Their experiments utilizing mice showed that the polysaccharide beta-glucan contained in Agaricus significantly activated the immune system. |
GASTRONOMIC EFFECTS
Agaricus Blazei Murill strengthens the immune function of inhibition for carcinomatous growth and activates natural killer cells. Among other components in Agaricus, the known anti-tumor agents are Beta-glucan and other polysaccharides which are highly concentrated in the powdered extract of Agaricus.
Polysaccharide contained in Agaricus Blazei Murill vitalizes generation of interferon and interleukin in guinea pigs. This effect indirectly functions to hold down proliferation of cancer cells. Also, it prevents viruses and other external matters from invading the tissue.
Clinical findings obtained jointly with universtity and hospital researchers were announced at the General Convention of the Japan Cancer Association in 1980. They showed that polysaccharide of Agaricus Blazei Murill is effective against Ehrlich's ascites carcinoma sigmoid coloniccancer, ovarian cancer, breast cancer, lung cancer and liver cancer as well as against solid cancer.
The findings also show that it activates metabolism by revitalizing normal biological tissue. Digestive enzymes such as amylase trypsin martase and the protease contained in fungus also enhances digestion. Besides, tyrosinase which oxydizes tyrosine and produces melanin, has a hypotensive effect. Furthermore, melanin protein that is effective in the generation of semen, hair and egg white was just recently discovered in Agaricus Blazei Murill.
Its physiological effects are anti-tumor, anti-allergic action and reduction of blood sugar level.

SKIN & MOISTURIZING
Functions of skin:
(1) to prevent evaporation of the moisture from the body.
(2) to defense against external irritations and invasion of bacteria, etc.
Skin Structure and Moisturizing:
The skin is roughly classified into epidermis, corium and hypoderm. The epidermis consists of horny layer (=the outermost layer), granular layer, spinous layer and basal layer. In particular, the horny layer in the epidermis is largely related to the flexibility of the healthy skin. The flexibility (or moisturizing) of the skin is maintained by the moisture contained in this horny layer.
Horny Layer Components and Moisturizing:
"Ceramides" are largely contained in the lipids constituting the horny layer. Namely, ceramides amount to approx. 40% of the intercellular lipids in the human horny layer, and play the major part in moisturizing the human skin.
CERAMIDES & GLYCOLIPIDS
Ceramides:
In general, a ceramide has a structure in which a fatty acid is bonded to sphingosine. In ceramides originating in wheat, sugars like mannose and glucose are further bonded to glycolipids. In human and animal bodies, ceramides are generated in the skin, blood, brain, spinal cord and nerve tissue, whereas in plants, they are generated in wheat, rice, soybean, maze, spinach, etc.
Glycolipids:
Substances containing both of sugar and lipid in a molecule are generally called glycolipids. Glycolipids are widely found in animal and plant bodies including human body and are regarded as essentially necessary in the activities of living organisms because of being important constituents of biomembranes.
FUNCTIONS OF CERAMIDES
Relief of Dry Skin:
Normal human horny layer contains from 10 to 30% moisture. When the moisture content in the horny layer is lowered to 10% or below due to some external reasons, the skin becomes less elastic, coarse and chapped. Such conditions may be relieved not merely by applying fats or oils to the skin but by feeding something capable of maintaining the moisture in the horny layer.
Relief of Aging & Atopic Dermatitis:
It is reported that ceramides contained in the horny layer are decreased when skin becomes more and more dried up through the progress of aging or the onset of atopic dermatitis.
Comparison with other Moisturizing components:
(1) Collagen and moisturizing: It is said that collagen which is a structural protein amounting to 70% of the corium, contributes tothe "tension" and elasticity of the skin.
(2) Hyaluronic acid and moisturizing: Hyaluronic acid which is known as a water soluble component of the skin, has been widely employed as a moisturizer in cosmetics.
(3) Ceramides and moisturizing: Recently, it is reported that not only water soluble moisturizing components but also intercellular lipids (i.e. ceramides) play important roles in moisturizing the skin.
CHARACTERISTICS OF "WHEAT GERM OIL-S"
Form:
It is in the form of a brown, viscous oil having the characteristic smell of germ oil.
Merits:
(1) It is completely made from wheat produced in Japan.
(2) It contains at least 6% of glycolipids including ceramides.
** The content of the ceramides in the glycolipids is not specified because no method hs been established for analyzing the content.
(3) It contains wheat germ oil as the major component (content: approx. 90%)
Functions:
(1) Moisturizing: When the moisture contnet in the human horny layer is determined by using a high frequency transmission moisture meter, it is found that an obvious moisturizing effect can be achieved by applying this product.
(2) Inhibition of collagenase: Collagen which is the major component of the corium is decomposed by an enzyme called collagenase. By adding this product, the collagenase activity is inhibited and thus collagen is stabilized.
(3) Suppression of POV(peroxide value): It is confirmed by a model experiment that an increase in POV can be suppressed by adding this product to foods and cosmetics.

Chitosan is a deacetylated derivative of chitin. It is polysaccharide found in nature with an amino group, contained in the shells of Crustacea such as crab, shrimp, etc. Chitosan is a white to light-red solid powder, insoluble in water, soluble in organic acid, but indigestible by human digestive enzymes. Therefore, it is one of the so-called dietary fibers.
The chemical structures of chitin, chitosan and glucosamine are shown below.
Crab shells are treated with dilute hydrochloric acid and dilute sodium hydroxide to remove calcium and protein. The product thus obtained is called chitin, and its further deacetylated product with concentrated sodium hydroxide is chitosan. Both chitin and chitosan are white to light-yellow or light-red substances with chemical structures quite similar to that of cellulose. Both are stable natural high molecular substances with strong inter-molecular binding force due to intramolecular amino group.
Glucosamine, a monosaccharide and component sugar of chitosan, can also be obtained by direct hydrolysis of chitin.
CHITOSAN AS FOOD MATERIAL
(1) It is a nitrogen-containing cationic polysaccharide found in nature, and is one of the water-soluble dietary fibers.

What is Perilla Seed ? Perilla frutescens and a lot of herbs like lavandula belong to Fam. LABIATAE. Perilla seed has been cultivated in Japan, China, Korea and Southeast Asia and used as spices and also main supply for ż-linolenate rich oil.
The patients with allergy are increasing every year, and the diet is said to be one of the most important factor of this disease. Daily intake of n-6 fatty acids, contained in meet or plant oil such as safflower, are accumulated as arachidonic acid which is converted to allergic mediators. PERILLA SEED EXTRACT inhibits the production of allergic mediators, termed leukotrienes (LTs). The mediators are metabolites of arachidonic acid through 5-lipoxygenase (5-LO) pathway. PERILLA SEED EXTRACT selectively inhibits 5-LO activity (in vitro). PERILLA SEED EXTRACT also inhibits type IV allergy as well as TPA-induced inflammation (in vivo). Moreover, the serum IgE levels were suppressed by administration of PERILLA SEED EXTRACT, and allergic rhinitis, hay fever and urticaria were significantly improved on human.

Product Standards for Perilla Leaf Extract Powder
MATERIAL
The material used is limited to Perilla leaf from Hokkaido (Japan) from the view point of its quality, its anti-allergic effects and the control of its production. The variety used in the manufacturing process is Perilla frutescens var. acuta Kudo forma viridis Makino.
EXTRACTION OF PERILLA LEAF
The decoction of the dried leaves afforded by boiling water gives an extract with greater anti-allergy activity than that obtained using aqueous ethanol and avoids the precipitation problems caused by chlorophyll etc., in the aqueous ethanol extract (Kosuna et al., 1994).
SPRAY DRYING
By the addition of dextrin to the aqueous Perilla extract, followed by spray drying, a stable powdered form is achieved, namely, Perilla extract powder with 1g equivalent to 1ml of Perilla extract. This powder is available for capsule and tablet formulations and for use in a range of health products.
